Acknowledgements First I would like to acknowledge my lab mates Allison Vestaal-Laborde, Andrew Eschenroder and Manisha Magar, without whom I would have been lost in my journey through the creation of this thesis. More importantly they were always incredibly good at keeping me laughing through all the pitfalls encountered in gathering my data and trying to put it to paper. giving me guidance and reassurance in structuring this project. My apprehensions about my data and conclusions were obliterated on our first meeting, and they proposed excellent follow-up experiments that made my isolated data fit into a wider scientific context. They have made this thesis a project I take pride in. Dr. Carmen Sato-Bigbee has been an incredible advisor during the last year. Taking a grad student into the lab seems like a daunting prospect, even in the best circumstances. With no lab experience and only a year to complete this thesis, her willingness to accept this challenge was truly inspiring. Carmen relentlessly challenged my assumptions about me and my thought process, forcing me to grow as a scientist and as a human being. Despite her relentless refrain of, " I'm just doing my job, " I know her passion and zeal are far beyond the level expected of an advisor, and I feel privileged to have worked with her. made possible my journey back into academia. They accepted me into their family when I was a young man adrift in a sea of self-bewilderment, and gave me the guidance to see that life isn't necessarily as bad or as complicated as I feared. Most importantly they gave me their amazing friendship, and to them I will always be eternally grateful. iv I dedicate this thesis to Barbara Schlegel-Jones. Our friendship in the last years of her life changed mine. Four years ago she remained convinced, despite my protestations, that I could become someone who would fulfill his potential. I will never forget her courage, strength, humor and wisdom.

اثر ویتامین B12 بر بیان Olig2 و غلظت کل پروتئین در کورتکس مغز موش
Background and purpose: Myelin-producing oligodendrocytes play a key role in supporting normal neuronal function of the mammalian central nervous system (CNS). Formation of myelinating oligodendrocytes from their precursors requires activation and coordination of a set of stage-specific transcriptional regulators that are important for the biosynthesis of myelin components. Olig2 plays an impor...
متن کاملMeCP2 Deficiency in Neuroglia: New Progress in the Pathogenesis of Rett Syndrome
Rett syndrome (RTT) is an X-linked neurodevelopmental disease predominantly caused by mutations of the methyl-CpG-binding protein 2 (MeCP2) gene. Generally, RTT has been attributed to neuron-centric dysfunction. However, increasing evidence has shown that glial abnormalities are also involved in the pathogenesis of RTT. Mice that are MeCP2-null specifically in glial cells showed similar behavio...
متن کاملNecl-4/SynCAM-4 Is Expressed in Myelinating Oligodendrocytes but Not Required for Axonal Myelination
The timing and progression of axonal myelination are precisely controlled by intercellular interactions between neurons and glia in development. Previous in vitro studies demonstrated that Nectin like 4 (Necl-4, also known as cell adhesion molecule Cadm-4 or SynCAM-4) plays an essential role in axonal myelination by Schwann cells in the peripheral nervous system (PNS). However, the role of Necl...
متن کاملThe Role of Endothelin Receptor A during Myelination of Developing Oligodendrocytes
Endothelin (ET)-1 and its receptors (ETA and ETB receptor) are present in the central nervous system. ET exerts biological effects on gliogenesis and glial cell functions. In order to define a possible mechanism of ETA receptor signaling, the distribution of the ETA receptor in developing oligodendrocytes and the effects of ET-1 on the myelination of oligodendrocytes were examined. ETA receptor...
متن کاملA New Player in CNS Myelination
The formation of the myelin sheath in the CNS is the endpoint of a defined developmental program along which oligodendrocytes progress. However, the molecular signals required for the initiation of myelination are largely unknown. Ishibashi et al. report in this issue of Neuron that ATP released by axons as a result of electrical stimulation serves as an important myelination signal. Surprising...
متن کاملA critical role for dorsal progenitors in cortical myelination.
Much controversy regarding the anatomical sources of oligodendrocytes in the spinal cord and hindbrain has been resolved. However, the relative contribution of dorsal and ventral progenitors to myelination of the cortex is still a subject of debate. To assess the contribution of dorsal progenitors to cortical myelination, we ablated the basic helix-loop-helix transcription factor Olig2 in the d...
